Item A new experimental in vitro culture medium for cultivation of Leishmania species(American Society for Microbiology, 1997) Limoncu M.E.; Balcioglu I.C.; Yereli K.; Ozbel Y.; Ozbilgin A.A new liquid culture medium prepared with chemicals that can be obtained economically and commercially was tested in in vitro cultivation of Leishmania promastigotes to obtain a large number of organisms to use in serological studies. The number of Leishmania infantum and Leishmania tropica promastigotes taken from Novy-MacNeal-Nicolle (NNN) medium reached 1 x 107/ml at the end of the 8th day in our new medium, though in NNN medium the number of organisms reached only 5 x 106/ml. After 10 subsequent passages, the culture medium prepared was evaluated as being quite inexpensive, simple, and successful compared with other commercially available liquid culture media.Item Incidence of Trichomonas vaginalis among women having vaginal discharge, in Manisa, Turkey.(1997) Yereli K.; Balcioglu I.C.; Degerli K.; Ozbilgin A.; Daldal N.Trichomoniasis characterise with a foamy yellowish odorous discharge, is an infection that causes superficial defects and necrotic ulcers in mucosa, and is spreadable with sexual contact. In T. vaginalis infection, the diagnosis can be made after the examination of saline wet mount preparations, stained smears preparation and cultivation of the vaginal discharge, the urine and the materials obtained from prostate secretion in males. In the present study, vaginal discharge samples obtained from 207 patients applied to Celal Bayar University, Medical Faculty Research Hospital, Manisa with vaginal discharge complaints were examined by utilising wet mount preparations, Giemsa staining method and CPLM cultivation method. In 27 of them (13.1%) T. vaginalis was detected by both saline wet mount preparation and Giemsa staining method. But as a result of the cultivation in CPLM media of the vaginal discharge materials collected from infected samples reproduction was observed in only 21 of them (10.2%). A treatment of Secnidazole by 2 gr. unique dose was applied to infected patients and their spouses and a complete recovery was followed up both microscopically and clinically.; The incidence of Trichomonas vaginalis was investigated in 207 women 18-45 years of age who presented to Celal Bayar University Hospital in Manisa, Turkey, with vaginal discharge. In 27 women (13.1%), T. vaginalis was detected by both direct microscopy of saline wet mount preparations and the microscopic examination of Giemsa stained smears. After cultivation of the vaginal discharge samples in CPLM media, T. vaginalis reproduction was seen in 21 samples (10.2%). All women with T. vaginalis as well as their spouses were treated with 2 g of Secnidazole and complete recovery was observed at follow-up 1 week after treatment ended.Item Giardiasis treatment in Turkish children with a single dose of ornidazole(2002) Özbilgin A.; Ertan P.; Yereli K.; Tamay A.T.; Kurt Ö.; Degerli K.; Balcioglu I.C.; Ok Ü.Z.; Onag A.This study was designed to compare the treatment efficacy of single dose of ornidazole with 5 d treatments of ornidazole and metronidazole in children with giardiasis. 175 children, between 2 and 15 y old, whose stool samples were found to be positive for Giardia lamblia cysts and/or trophozoites by either saline-Lugol, formalin-ethyl acetate or trichrome staining, were enrolled in the study. Of these children, 105 were treated with a single dose of ornidazole: 35 with 30 mg/kg, 35 with 25 mg/kg and 35 with 20 mg/kg; 35 were treated with 25 mg/kg per day of ornidazole for 5 d in 2 doses and 35 children were treated with 20 mg/kg per day metronidazole for 7 d in 3 doses. All cases were examined on the 7th, 10th and 14th days after treatment by the same methods; clinical symptoms were also evaluated. Giardia lamblia was eradicated in 34 of 35 (97%), 34 of 35 (97%) and 33 of 35 (94%) patients treated with 30, 25 and 20 mg/kg single doses of ornidazole, respectively. Eradication was achieved in all 35 patients treated with 25 mg/kg per day ornidazole for 5 d and in 31 of 35 (89%) patients treated with metronidazole. There was no statistically significant difference among doses of ornidazole (p > 0.05); however, all ornidazole treatment regimens were significantly more effective than metronidazole treatment (p < 0.05). No important side-effects were detected in any patients and clinical symptoms disappeared in all. Single-dose ornidazole treatment could be considered as a proper and effective alternative method for the treatment of giardiasis in children.Item Effectiveness of peptone-yeast extract (P-Y) medium in the cultivation and isolation of Entamoeba histolytica/Entamoeba dispar in Turkish patients(2003) Dagci H.; Balcioglu I.C.; Ertabaklar H.; Kurt O.; Atambay M.Amebiasis is a common protozoan infection worldwide, causing serious health problems in both children and adults. Today, almost 10% of the world population is infected with Entamoeba histolytica/Entamoeba dispar. The aims of this study were both the comparison of the reproduction rates and densities of E. histolytica/E. dispar in Robinson, Dobell-Laidlaw and P-Y culture media and isolation of E. histolytica/E. dispar from stool samples in Peptone-Yeast (P-Y) medium. Trophozoites and cysts of E. histolytica/E. dispar, maintained in Robinson medium, and stool samples of patients with amebiasis were inoculated into P-Y, Robinson and Dobell-Laidlaw culture media. Reproduction rates reached their peak levels 48 h after the inoculation in all culture media. Reproduction rates in P-Y and Robinson media were found similar; however, they were higher than the reproduction rate in Dobell-Laidlaw medium (p < 0.01); there was no statistically significant difference between the reproduction rates of P-Y and Robinson media (p > 0.05). Twelve isolates from 12 patients were cultivated in P-Y medium and checked for reproduction everyday for 7 days. Twelve of the 12 (100%) isolates were cultivated in P-Y medium, indicating that the P-Y was an effective medium for the isolation of E. histolytica/E. dispar in stool samples. According to these results, P-Y medium could be preferred in immunologic, serologic and molecular studies and, thus the definitive diagnosis of amebiasis due to its low cost and simple formula. © 2003 Elsevier Science Inc. All rights reserved.Item Incidence of giardiasis among siblings in Turkey(2003) Balcioglu I.C.; Limoncu E.; Ertan P.; Yereli K.; Özbilgin A.; Onag A.Background: Giardiasis, a common infection among children, is caused by a flagellated protozoan called Giardia lamblia. It is well known to be contagious in common living places. This is an epidemiologic study investigating the incidence of giardiasis among the siblings of patients with giardiasis living in the same household. Methods: Stool samples of 50 patients with a G. lamblia positive sibling in the same household, and 50 patients with a G. lamblia negative sibling in the same household, were examined for giardiasis by saline-Lugol, formalin-ethyl acetate concentration and trichrome staining methods. Other causes of diarrhea were excluded by microbiologic laboratory tests. Results: Thirty-eight per cent of siblings of G. lamblia positive patients and 8% of siblings of G. lamblia negative children were found to be positive for G. lamblia cysts and/or trophozoites. The differences between the groups were statistically significant (P < 0.001). Conclusion: The results suggested that the examination of G. lamblia among the siblings of patients with giardiasis both in the same age group and living in the same household should be considered.Item Serological and entomological survey in a zoonotic visceral leishmaniasis focus of North Central Anatolia, Turkey: Corum province(Elsevier, 2005) Ertabaklar H.; Toz S.O.; Ozkan A.T.; Rastgeldi S.; Balcioglu I.C.; Ozbel Y.In the present study, we aimed to carry out an epidemiological and entomological survey on a visceral leishmaniasis (VL) focus located on the northern central part of Anatolia, Turkey. Five villages of Corum province, where five confirmed cases of human visceral leishmaniasis (HVL) (one patient/village) were reported between June 1998 and August 2001 were included in the study. A total of 625 children and 131 dogs were sampled and the physical examination was carried out by authorized physicians and veterinarians. An indirect fluorescent antibody test (IFAT) was performed by standard procedures for human and dog sera, while the direct agglutination test (DAT) was only performed for dog sera. Sand fly collection was performed in three villages by CDC miniature light traps. Hepatosplenomegaly and hepatomegaly were detected in two and eight children, respectively. The seropositivity rate among children was found to be 0.16% (1/625) in the region. The seroprevalence of canine infection in these five villages ranged between 0.0% and 28.26%. In two villages, named Ahlatcik and Asagifindikli, no seropositive dogs were found. A total of 1218 sand flies were collected throughout the study. Six species of Phlebotomus were identified: P. transcaucasicus, P. neglectus, P. halepensis, P. tobbi, P. papatasi, and P. jacusieli. P. transcaucasicus was found to be the predominant species in Cevizli (47.44%; 343/723) and Ucoluk (79.95%; 351/439) villages, while P. tobbi was abundant in Kucukerikli (42.85%; 24/56). © 2005 Elsevier B.V. All rights reserved.Item Rural life, lower socioeconomic status and parasitic infections(2007) Balcioglu I.C.; Kurt Ö.; Limoncu M.E.; Dinç G.; Gümüş M.; Kilimcioglu A.A.; Kayran E.; Özbilgin A.This study was conducted both to assess the prevalence of parasitic infections in two neighboring villages of Manisa province in Turkey, with different socioeconomic levels and investigate the influences of some individual and environmental factors on these infections. A total of 100 school children were enrolled and their cellophane tape and stool samples were examined for parasites. Pediculus capitis infestation was diagnosed by visual inspection of the hair. Although the prevalences of both intestinal parasitosis and pediculosis were high in both villages, there was a statistically significant difference for pediculosis, but not for intestinal parasitosis. Lack of social security and father's regular job were found to be highly correlated with all parasitic infections. Pediculosis was more common in younger children, girls and those in crowded families. These results indicate not only the high prevalence of parasitic infections, but also the urgent need for the improvement of basic health services and infrastructure in the rural areas, crowded with poorer immigrant population. © 2007 Elsevier Ireland Ltd. All rights reserved.Item Short report: Quantiferon-Leishmania as an epidemiological tool for evaluating the exposure to Leishmania infection(2010) Turgay N.; Balcioglu I.C.; Toz S.O.; Ozbel Y.; Jones S.L.The aim of the present preliminary study was to investigate the potential of measurement of IFN-γ secretion by T cells into blood plasma using QuantiFERON assay with leishmanial antigens to determine the presence of Leishmania infection. Blood samples from cured visceral (N = 18), and cutaneous (N = 20) leishmaniasis cases, and 20 healthy controls were tested. The IFN-γ responses to Leishmania major H2B and Leishmania infantum H2B antigens were detected from the majority of treated old visceral leishmaniasis cases, but not from controls. Future studies using larger groups will be required to establish the true potential of the assay for epidemiological screening of leishmaniasis. Twenty-one VL cases were reported within six years from settlements located in most northeastern Turkey and we therefore aimed to carry out an entomological and seroepidemiological survey in this new focus for clarifying risk factors. Blood samples from 290 children and 165 dogs were collected. Sera samples were investigated for anti- Leishmania antibodies using indirect fluorescent antibody test. Sand fly collection for determining the fauna and seasonal activity was performed in all settlements by CDC light traps between June and September 2006. Although no seropositive child was detected during the survey the overall seroprevalence rate of canine leishmaniasis was found as 7.2%. A total of 4154 sand flies were collected and 10 species of genus Phlebotomus were identified belonging to A. dlerius, Larroussius, Paraphlebotomus and Phlebotomus subgenera. Among them Phlebotomus kandelakii s.l. (55.44%), Phlebotomus balcanicus (12.62%) and Phlebotomus neglectus (4.40%) was detected as probable vector species for this new focus. The poor sanitation, very high population size of sand flies, probably because of very short season, no control measures for sand flies as well as dogs, and presence of microclimate suitable for sand flies were considered as main risk factors in the area. © 2015 Elsevier B.V.Item The Toxicity of Essential Oils From Three Origanum Species Against Head Louse, Pediculus humanus capitis(Springer Science and Business Media Deutschland GmbH, 2021) Arserim S.K.; Cetin H.; Yildirim A.; Limoncu M.E.; Cinbilgel I.; Kaya T.; Ozbel Y.; Balcioglu I.C.Purpose: Pediculosis caused by human head louse, Pediculus humanus capitis De Geer (Anoplura: Pediculidae) is a public health problem with a high prevalence, especially among children in school age. Different chemical pediculicides (insecticides) are used against lice but the repeated application of these pediculicides for the treatment of P. humanus capitis infestation in the past decades has resulted in the development of resistance. The plant essential oils have been used as alternative sources of lice treatment agents. Pediculicidal activity of the essential oils of three Origanum species (O. onites, O. majorana and O. minutiflorum) against P. humanus capitis adults was examined in this study. Methods: Essential oils from aerial parts of three Origanum species were obtained by hydrodistillation using a Clevenger-type apparatus. P. humanus capitis specimens used were obtained from infested children by combing method at primary schools. Adult lice were exposed to three dilutions (1%, 0.5% and 0.1% w/v) of tested oils for 5 min using the adult immersion test. Results: According to the results, all Origanum essential oils at 1% concentration have been shown to significantly decrease the rate of limb, bowel and abdomen movements, producing more than 90% mortality after 12 h. Conclusion: Our results indicated that Origanum essential oils can be used for the development of new pediculicides against head louse. © 2021, Witold Stefański Institute of Parasitology, Polish Academy of Sciences.Item Analysis of morphological variations of three Adlerius (Diptera: Psychodidae) species collected in two cutaneous leishmaniasis endemic foci of Turkey(Springer Science and Business Media Deutschland GmbH, 2022) Kavur H.; Arikan H.; Toz S.; Balcioglu I.C.; Ozbel Y.Leishmaniasis, transmitted to human by sand flies, is a neglected vector-borne diseases and has two clinical forms: cutaneous (CL) and visceral leishmaniasis (VL) in Turkey. Subgenus of Adlerius are known as probable vectors of both forms of leishmaniasis in Turkey. Examined three species belonging to Adlerius are difficult to distinguish morphologically and have similar taxonomic characters. The aim of this study was to analyse and identify the possible morphological variations, of wild-caught sand fly specimens belonging to Adlerius subgenus. Six and seven morphological characters for female and male specimens were used, respectively. ANOVA, principal component analysis (PCA) and discriminant function analysis (DFA) test were performed to determine the significant morphometric variables of the three species. Univariate and multivariate analyses of 13 morphometric variables via ANOVA, DFA and PCA showed that eight (six for females, two for males) morphometric variables had an accuracy of 100% for discriminating three Adlerius subgenus specimens. The analyses revealed significant differences on several morphological structures of Adlerius specimens. These results suggested that morphological and morphometrical features can be used to discriminate three Adlerius species, in which one of them is proven vector Leishmania (L.) tropica in two foci of Turkey. © 2021, African Association of Insect Scientists.
