Item Karyotype, C- and G-band Patterns and DNA content of Callimenus (=Bradyporus) macrogaster macrogaster(Library of the University of Arizona, 2002) Turkoglu S.; Koca S.Chromosomes and detailed karyotype information (the number, shape, relative length, arm ratio, centromeric index) of Callimenus (=Bradyporus) macrogaster macrogaster Lef. (Orthoptera: Tettigonioidea, Bradyporini) of Turkey belonging to the subfamily Bradyporinae are described. The diploid number of chromosomes was found to be 2n ♂= 23, with 2 metacentric pairs, 2 submetacentric pairs, 6 acrocentric pairs. The X chromosome is metacentric. This species has a XX ♀/X0 ♂ sex determining mechanism. The basic karyotype is complemented by a description of C- and G-banding patterns. The C-banding pattern in spermatogonial metaphase was characterized by the presence of paracentromeric C-bands in all chromosomes, and a distal C-band in chromosome 5. The G-banding pattern was complex. The 2C nuclear DNA content was found to be 10.26 ± 0.16 picograms by microspectrophotometry.Item Investigation of histopathological and cytogenetic effects on Lepomis gibbosus (Pisces: Perciformes) in the çine stream (Aydin/Turkey) with determination of water pollution(2005) Koca Y.B.; Koca S.; Yildiz Ş.; Gürcü B.; Osanç E.; Tunçbaş O.; Aksoy G.Water quality and the distribution of some heavy metals in three different organs of Lepomis gibbosus from the Çine Stream were studied. Also, histopathological changes in gill, liver, and muscle tissue were examined at light microscopical level. Micronucleus (MN) formation in fish erytrocytes, as an indicator of chromosomal damage, has been increasingly used to detect the genotoxic potential of environmental contaminants. The frequency of MN was examined from samples of fish from the Çine Stream and a control group. MN frequency was higher in fish samples caught from the Çine Stream than that in the control group. The chemicals ammonia, nitrite, nitrate, orthophosphate, and sulphate were determined as parameters that possibly affect the gill, liver, and muscle morphology. Zn was the most accumulated metal in tissues as well as in water. Maximum metal accumulation occured in both liver and gills. For histopathological examinations, samples of gills, liver, and muscle tissues of L. gibbosus were studied by using light microscopy. In this study, a significant decrease in mean length of primary and secondary lamellae were observed. Moreover, cellular proliferation developed with secondary lamellae fusion, ballooning degenerations or club deformation of secondary lamellae, as well as distribution of necrotic, hyperplastic and clavate secondary lamellae. In the liver, altered staining, swollen and ruptured parenchymal cells, loss of cord structure, reduce of glycogen in hepatocytes, and vacuolar structure filled with cellular debris and many dark particles were seen. In muscle tissue, focal necrosis, cellular dissolution, and a decline or loss of striatation in muscle fibres were found. © 2005 Wiley Periodicals, Inc.Item A cytogenetic study on the some Western Anatolian Triturus species (Salamandridae, Urodela); [Bazı batı anadolu Triturus türleri (Salamandridae, Urodela) üzerinde sitogenetik bir çalışma](Veteriner Fakultesi Dergisi, 2008) Özdal Kurt F.; Koca S.The karyotypes of two species in the name of Triturus vulgaris and Triturus karelinii, belonging Salamandridae family of Urodela order were analyzed. Chromosome numbers of both the Triturus species were identified as 2n=24 and chromosomes forming karyotypes clustred in three group, long, medium and short by their lengths. I, VIII and XIIth chromosome pairs in Triturus vulgaris showed metacentric morphology whereas the same chromosomes showed submetasentric morphology in Triturus karelinii. VI, VII, IX, X and XIth chromosome pairs in Triturus vulgaris were found to be submetasentric compared to metasentric morphology of same chromosome pairs in Triturus karelinii. The other chromosome pairs II, III, IV and Vth were found to have metasentric morphology in both species. According to our measurements, chromosomes of Triturus karelinii were found to be long and to have more regular line and intrakaryotypic variations between chromosomes were determined as low.Item Patterns of Care for Lung Cancer in Radiation Oncology Departments of Turkey(2008) Demiral A.N.; Alicikus Z.A.; Işil Ugur V.; Karadogan I.; Yöney A.; Andrieu M.N.; Yalman D.; Pak Y.; Aksu G.; Özyigit G.; Özkan L.; Kilçiksiz S.; Koca S.; Çaloǧlu M.; Yavuz A.A.; Başak Çaǧlar H.; Beyzadeoǧlu M.; Iǧdem S.; Serin M.; Kaplan B.; Koç M.; Korkmaz E.; Karakoyun-Çelik O.; Dinçer S.; Kinay M.Purpose: To determine the patterns of care for lung cancer in Turkish radiation oncology centers. Methods and Materials: Questionnaire forms from 21 of 24 (87.5%) centers that responded were evaluated. Results: The most frequent histology was non-small cell lung cancer (NSCLC) (81%). The most common postoperative radiotherapy (RT) indications were close/(+) surgical margins (95%) and presence of pN2 disease (91%). The most common indications for postoperative chemotherapy (CHT) were "≥ IB" disease (19%) and the presence of pN2 disease (19%). In Stage IIIA potentially resectable NSCLC, the most frequent treatment approach was neoadjuvant concomitant chemoradiotherapy (CHRT) (57%). In Stage IIIA unresectable and Stage IIIB disease, the most frequent approach was definitive concomitant CHRT (91%). In limited SCLC, the most common treatment approach was concomitant CHRT with cisplatin+etoposide for cycles 1-3, completion of CHT to cycles 4-6, and finally prophylactic cranial irradiation in patients with complete response (71%). Six cycles of cisplatin + etoposide CHT and palliative thoracic RT, when required, was the most commonly used treatment (81%) in extensive SCLC. Sixty-two percent of centers did not have endobronchial brachytherapy (EBB) facilities. Conclusion: There is great variation in diagnostic testing, treatment strategies, indications for postoperative RT and CHT, RT features, and EBB availability for LC cases. To establish standards, national guidelines should be prepared using a multidisciplinary approach. © 2008 Elsevier Inc. All rights reserved.Item Genotoxic and histopathological effects of water pollution on two fish species, Barbus capito pectoralis and Chondrostoma nasus in the Büyük Menderes River, Turkey(2008) Koca S.; Koca Y.B.; Yildiz Ş.; Gürcü B.The genotoxic and histopathological effects of water pollution were investigated on two fish species caught from the Buyuk Menderes River and from its tributary, the Cine Stream. The Buyuk Menderes basin is an important agricultural area in Turkey. The levels of copper, zinc, cadmium, cobalt, and lead were measured at the surface of the water and in gills, liver, and muscle tissue of Chondrostoma nasus and Barbus capito pectoralis. In some tissues, the concentrations of some of these metals exceeded acceptable levels for human consumption. Zinc was found to be the most abundant metal in water and tissues. Maximal metal accumulation was observed in the liver. To detect the genotoxic potential of contaminants, the formation of micronucleus in erythrocytes was used as indicator of chromosomal damage. The frequency of micronucleus formation did not show significant differences between locations and controls in B. capito pectoralis caught from three locations and C. nasus from two locations. The histological changes included significant decreases of the mean lengths of primary and secondary lamellae. In gills epithelia, we observed cellular proliferation that developed Because of secondary lamellae fusion, ballooning degenerations, or club deformation of secondary lamellae and cystic structures in secondary lamellae. In the liver, the changes included swollen and ruptured parenchymal cells, loss of cord structure, vacuoles filled with cellular debris, focal necrosis, and a significant increase in Kupffer cells. © 2007 Humana Press Inc.Item Investigation of histopathological and cytogenetic effects of heavy metals pollution on Cyprinus carpio (Linneaus, 1758) in the Göhnarmara Lake, Turkey(2010) Gurcu B.; Yildiz S.; Koca Y.B.; Koca S.Concentration of heavy metals were measured in the surface water, sediments and three different organs of Cyprinus carpio from the Gölmarmara lake (Manisa, Turkey). Four sampling sites located at different parts of the lake were pre-defined. The accumulation of copper, zinc, cadmium, cobalt, lead, chromium, iron, mangane in gills, liver and muscle of C. carpio were determined. Also, histopathological changes in gill, liver and muscle tissue were examined at light microscopical level. The order of accumulation of heavy metals was found to be Zn>Pb>Fe>Co>Mn>Cr>Ni>Cu>Cd in water, Zn>Cr>Pb>Fe>Co>Mn>Ni>Cu>Cd in sediment, Zn>Cr>Pb>Ni>Cu>Fe>Co>Cd in muscle and Zn>Cr>Pb>Cd>Ni>Cu>Fe>Co in liver and Zn>Cr>Ni>Pb>Cu>Cd>Co>Fe in gills. As a result of histopathological examinations, a significant decrease in mean length of primary and secondary lamellae was observed. Cellular proliferation caused secondary lamellae fusion, ballooning degenerations of secondary lamellae as well as distribution of necrotic and clavate secondary lamellae. Water, sediment and fish tissue samples taken from different sites of 3 different study areas were studied and the order of accumulation of metals was Fe>Pb>Mn>Co>Zn>Ni> Cr>Cu>Cd in water, Fe>Co>Mn>Pb>Zn>Ni>Cr>Cu>Cd in sediment and Cd>Mn>Cu>Cr>Ni>Fe>Zn>Co>Pb, Cd>Pb>Ni>Cr>Mn>Cu>Fe>Co>Zn and Cr>Ni>Zn>Cd>Pb>Mn>Cu>Fe>Co in liver, muscle and gill of fish, respectively. In histopathological studies, a decrease in the length of primary and secondary lamellae of gills, fusion in secondary lamellae, cellular proliferation, clavate lamellae formation and necrosis were observed. In liver tissue, dilation of sinusoid, increase in the number of erythrocytes, ruptured hepatocytes, decrease in glycogen accumulation and vacuolization were observed. In muscle tissue, necrosis, cellular dissolution and loss of striatation in muscle fibers were found. It was observed that pollution of water had no genotoxic effect onAnguilla anguilla. © Medwell Journals, 2010.Item Comparison of survival with somatostatin analog and chemotherapy and prognostic factors for treatment in 165 advanced neuroendocrine tumor patients with Ki-67 20% or less(Lippincott Williams and Wilkins, 2016) Özaslan E.; Karaca H.; Koca S.; Sevinc A.; Hacioglu B.; Özkan M.; Özcelik M.; Duran A.O.; Hacibekiroglu I.; Yildiz Y.; Tanriverdi O.; Menekse S.; Aksoy A.; Bozkurt O.; Urvay S.; Uysal M.; Demir H.; Ciltas A.; Dane F.The objectives of this study were to compare progression-free survival (PFS) with somatostatin analog (SSA) versus chemotherapy (CTx) in first-line therapy and to determine the patient group in which these treatments were more effective in neuroendocrine tumors (NETs) with a Ki-67 index of 20% or less. Patients who received SSA or CTx and had unresectable locally advanced and metastatic NETs with a Ki-67 index of 20% or less were retrospectively selected from 13 centers in the Turkish database between 2000 and 2015. One hundred and sixty-five patients were enrolled. The median age was 56 years and the male-to-female ratio was 1.09. Seventy-four (45%) patients were of grade 1 NET and 91 (55%) were of grade 2. SSA was given to 104 patients, whereas 61 were treated with CTx. The objective response rate after SSA was 15.4%; another 73.1% had stable disease. The objective response rate after CTx was 36.1%, and 40.9% had stable disease (P=0.008). The median PFS in SSA patients was 21 months (95% confidence interval: 12.4-29.6), and 8 months for CTx (95% confidence interval: 5.5-10.6) (P<0.001). There was no significant difference between PFS of receiving SSA and CTx in pancreatic neuroendocrine tumor (PNET) patients; however, the PFS of receiving SSA was longer in non-PNET patients (P<0.001). SSA was better treatment in advanced NET patients with a Ki-67 index of less than 5%, having a primary resected and a performance status of 0 (P<0.05). The purpose of this study was to evaluate the clinicopathologic features and their impact on outcomes specifically in patients diagnosed with HGUPS. In this multicenter trial, we retrospectively analyzed 112 patients who were diagnosed and treated at 12 different institutions in Turkey. We collected data concerning the patients, tumor characteristics, and treatment modalities. There were 69 males (61.6 %) and 43 females (38.4 %). Median age was 56 years (19–90). The most common anatomic site of tumor origin was the upper extremity. Pleomorphic variant was the predominant histological subtype. Median tumor size was 8.2 cm (0.6–30 cm). Tumors were mainly deeply seated (57.1 %). Fifty-seven patients (50.9 %) were stage II and the remainder were stage III at the time of diagnosis. Median follow-up was 30 months (2–160). The primary site of distant metastasis was the lung (73.5 %) and the second most common site was the liver (11.7 %). The 5-year overall survival, distant metastasis-free survival, and local recurrence-free survival rates were 56.3, 53.4, and 67.2 %, respectively. Multivariate analysis showed that Eastern Cooperative Oncology Group (ECOG) performance score of II (p = 0.033), deep tumor location (p = 0.000), and development of distant metastasis (p = 0.004) were negatively correlated with overall survival, and perioperative radiotherapy and negative microscopic margins were significant factors for local control rates (p = 0.000 for each). Deep tumor location (p = 0.003) was the only adverse factor related to distant metastasis-free survival. Deep tumor location, ECOG performance score of II, and development of distant metastasis carry a poor prognostic implication on overall survival. Sixteen potential prognostic variables were assessed for analysis. Results: The patients had received a median of 5 prior chemotherapy regimens and a median of 3 eribulin cycles for MBC. Median progression-free survival (PFS) was 5.5 months (95% Cl: 4.1-7.8) and median overall survival (OS) was 11 months (95 % Cl: 6-15). Multivariate analysis showed that eribulin treatment line was shown to have independent prognostic significance for PFS. PFS difference was demostrated in patients who received 3 chemotherapy lines for advanced disease compared to those who had more than 3 chemotherapy lines [median PFS; 3 lines: 8.6 months (6.2-11) and >3 lines: 4.6 months (3.7-4.6) p=0.00]. The clinical benefit rate (CBR) was 52.5 and 35% in patients treated with three lines and with >3 previous chemotherapeutic regimens. Most common toxicities were neutropenia (62.5%), fatigue (52.5%), alopecia (50%) and nausea (37.5%). All rights reserved.Item Genotoxic Effect, Oxidative Stress and Cell Death due to Metronidazole Application in Gills and Liver Tissues of Rainbow Trout (Oncorhynchus mykiss)(University of Tehran, 2022) Gurcu B.; Koca S.; Koca Y.B.; Collu F.; Ibrahim Tuglu M.In this study, the purpose was to investigate the histopathological, genotoxic effect, oxidative stress and cell death due to Metronidazole (MTZ), which is a 5-nitroimidazole compound, used widely for the treatment of anaerobic organism infections in fish and humans on gill and liver tissues of Oncorhynchus mykiss. Trout fishes were exposed to 5, 10, and 20 mg/L of MTZ in the aquariums for 2, 4 and 8 days. Staining technics namely H&E, NOS immunohistochemistry, and TUNEL were performed to determine histopathological changes, oxidative damage and apoptosis. Additionally, smear preparations were also prepared from gill blood for genotoxic evaluations. The organ damage started in the 2nd day with 5 mg/L MTZ application and effects increased per duration and dose-dependent manner. It was observed that the gills had the primary and secondary lamellae lengths, with formation of clavate lamellae, fusion in secondary lamellae, separation of epithelium and aneurysm. Regional necrosis, vacuolization of hepatocytes, pycnotic nucleus, enlarged sinusoids were also determined in the liver. NOS immunoreactivity increased with the inducible immunoreactivity (iNOS) that was more prominent when compared to the endothelial immunoreactivity (eNOS). Apoptotic immunoreactivity was higher in the 10 mg 8th day experimental group at liver and gills, and was lower 20 mg 8th day experimental group. When the gills and liver compared with each other, in all doses, immunoreactivity was lower in gills, compared with liver. Genotoxic examinations showed that both number of micro nucleated erythrocytes and nuclei abnormalities were higher in MTZ-treated groups. © 2022 University of Tehran. We aimed to investigate the efficacy and safety of second-line FOLFIRI and aflibercept combination in patients with metastatic CRC in real-life experience. Materials and Methods: Four hundred and thirty-three patients who treated with FOLFIRI and aflibercept in the second-line were included in the study. The clinical and pathological features of the patients were recorded retrospectively. Survival (overall and progression-free survival [PFS]), response rates, and safety data were analyzed. Results: The median age was 61. Majority of patients (87.5%) received first-line bevacizumab and 10.1% of patients received anti-epidermal growth factor receptor agents. About 80% of patients had KRAS, 18.6% of patients had NRAS, and 6.4% of patients had BRAF mutations. The median OS was 11.6 months (95% confidence interval [CI], 10.6-12.6) and the median PFS was 6 months (95% CI, 5.5-6.5). About 4.6% of patients had complete response and 30.6% of patients had partial response as best tumor response. Grade 1-2 toxicities were seen in 33.4% of patients, while grade 3-4 toxicities were recorded in 27% of patients. Eight patients (2%) died due to treatment toxicity. Conclusions: Overall and PFS were similar in routine clinical practice compared to phase III pivotal VELOUR trial. However, response rates were found to be higher. It was observed that there were fewer adverse events compared to the VELOUR trial. © 2022 Authors. All rights reserved.Item Determination of histopathological effects and myoglobin, periostin gene-protein expression levels in Danio rerio muscle tissue after acaricide yoksorrun-5EC (hexythiazox) application(Taylor and Francis Ltd., 2023) Başımoğlu Koca Y.; Koca S.; Öztel Z.; Balcan E.Although pesticides are essential agrochemicals to annihilate harmful organisms in agriculture, their uncontrolled use has become an important threat to environmental health. Exposure to pesticides can affect many biological systems including immune system, endocrine system, and nervous system. However, the potential side effects of pesticides to skeletal muscle system remain unclear. Present study has focused on the evaluation of this issue by using an acaricide, yoksorrun-5EC (hexythiazox), in an aquatic model organism, Danio rerio. The histological analyses revealed that increased concentrations of the acaricide cause degradation of skeletal muscle along with increased necrosis and atrophy in myocytes, intercellular edema, and increased infiltrations between perimysium sheaths of muscle fibers. The effects of acaricide on myoglobin and periostin, which are associated with oxygen transport and muscle regeneration, respectively, were investigated at the gene and protein levels. RT-PCR results suggested that high concentration yoksorrun-5EC (hexythiazox) can induce myoglobin and periostin genes. Similar results were also obtained in the protein levels of these genes by western blotting analysis. This retrospective study aimed to assess the activity and tolerability of Lutetium-177 (Lu-177) PSMA-617 radioligand therapy (RLT) in mCRPC patients across multiple cancer centers in Turkey. The study included 165 patients who received at least one cycle of Lu-177 PSMA-617 RLT, with the majority having bone metastases and undergone prior treatments. Prostate-specific antigen (PSA) levels were assessed before each treatment cycle, and the biochemical response was evaluated in accordance with the Prostate Cancer Work Group 3 Criteria. The PSA decline of ≥50% was classified as a response, while an increase of ≥25% in PSA levels was indicative of progressive disease. Neither response nor progression was considered as stable disease. The Lu-177 PSMA-617 RLT led to a significant PSA response, with 50.6% of patients achieving a >50% decrease in PSA levels. Median overall survival (OS) and progression-free survival were 13.5 and 8.2 months, respectively. Patients receiving Lu-177 PSMA-617 RLT in combination with androgen receptor pathway inhibitors (ARPIs) had a higher OS compared to those receiving Lu-177 PSMA-617 RLT alone (18.2 vs 12.3 months, P =.265). The treatment was generally well-tolerated, with manageable side effects such as anemia and thrombocytopenia. This study provides real-world evidence supporting the effectiveness and safety of Lu-177 PSMA-617 RLT in mCRPC patients, particularly when used in combination with ARPIs. These findings contribute to the growing body of evidence on the potential benefits of PSMA-targeted therapies in advanced prostate cancer. © 2023 UICC.
