Item Which one is the predictor of carotid intima media thickness in patients with glomerulonephritis; neutrophil-to-lymphocyte ratio or proteinuria?; [Glomerülonefritli hastalarda karotis intima media kalınlığının prediktörü hangisidir; nötrofil-lenfosit oranı mı proteinüri mi?](Pamukkale University, 2019) Toraman A.; Eren B.; Düzgün F.; Taneli F.; Kürşat S.Purpose: Glomerulonephritis is inflammation of the glomeruli and proteinuria itself is a reflection of the glomerular inflammation. In this study we aimed to investigate the relationship between carotid intima media thickness (CIMT) one of the earliest findings of atherosclerosis and markers of glomerular inflammation. Materials and Methods:40 patients with biopsy proven glomerulonephritis and 46 healthy controls were enrolled in the study. Human Growth arrest spesific protein 6 (Gas6) levels in serum samples were studied by ELISA. CIMT measurement was performed by the same radiologist. Neutrophil-lymphocyte ratio (NLR) was calculated by dividing the number of neutrophils by the number of lymphocytes. Results: The mean age was 42.88±15.41 in patient group and 38.26±9.04 in controls. The mean duration of illness was 29.07±52.90 mounts, proteinuria was 4027.05±4030.22 mg/day, Modification of Diet in Renal Diseases Study Glomerular Filtration Rate (MDRD-GFR) was 53.80±48.32 mL/min/1.73m2. CIMT was 0.62±0.17 mm in patient group, 0.46±0.10 mm in controls. Neutrophil-to-Lymphocyte Ratio (NLR) was 3.69±4.46 in patient group and significantly higher than control group (1.74±0.63). Gas6 levels were statistically higher in control group. CIMT was positively correlated with age, fibrinogen, ferritin, proteinuria and NLR and negatively correlated with HDL cholesterol and Gas6 in glomerulonephritis group. Age was the predictor for CIMT in the logistic regression model. In all group CIMT was positively correlated with age, creatinine, uric acid, fibrinogen, ferritin, CaxP product, proteinuria and NLR and negatively correlated with hemoglobin level, Gas6. In lineer logistic regression analysis carotid IMT was significantly associated with age and ferritin and proteinuria. Conclusion: We have shown that proteinuria is one of the main determinants of increased CIMT independent from the GFR levels in glomerulonephritis with relatively preserved glomerular filtration rate. © 2019, Pamukkale University. All rights reserved.Item The effect of DEBIO 1143 usage alone or in combination with tamoxifen on estrogen receptor positive breast cancer cell lines; [DEBIO 1143’ün tek başına veya tamoxifenle kombinasyonunun östrojen reseptörü pozitif meme kanseri hücre hatlarındaki etkisi](Pamukkale University, 2020) Tuğrul B.; İşseven M.Purpose: To investigate the effect of increased concentrations of tamoxifen (TAM) and DEBIO 1143 (AT-406) administered alone or in combination on cells in MCF-7 and BT-474 estrogen receptor positive (ER+) breast cancer cell lines. Materials and methods: The effect of tamoxifen and DEBIO 1143 administered alone or in combination on cell viability in MCF-7 and BT-474 cell lines at the 72nd hour was assessed by the XTT test. Multi-parameter apoptosis assay kit was used to display the effect of the alone/combination of TAM and DEBIO 1143 on both cell lines. Fluorescence microscobic analysis was performed. Results: The IC50 value of TAM was 3.8±0.6 micromolar (μΜ) and 18.9±6.7 μΜ in the MCF-7 and BT-474 cell lines, respectively. The IC50 value of DEBIO 1143 was 15±0.5 μM in the MCF-7 cell line. The results related to drug combination were statistically significant for both cell lines (p<0.001). The decrease in cell viability was not associated with apoptosis. Conclusion: In ER+breast cancer cell lines, the combined doses of the TAM and DEBIO 1143 reduced cell viability more than their administration alone. Combined administrations in both cell lines were concluded in a synergistic effect. Further research is needed to determine which cell death type other than apoptosis is associated with a reduction in cell viability caused by combined administration. © 2020, Pamukkale University. All rights reserved.Item Investigation of neuraminidase 1 gene association in Henoch-Schönlein Purpura (HSP) with renal involvement; [Henoch Schönlein purpura vaskülitinde nöraminidaz-1 geni ile böbrek tutulumunun ilişkisi](Pamukkale University, 2022) Yılmaz N.B.; Ertan P.; Yüksel S.; Neşe N.; Horasan G.D.; Berdeli A.H.Purpose: HSP is a common small vessel vasculitis. It is the most common cause of non-thrombocytopenic purpura in childhood. The role of genes in etiopathogenesis of the disease, which has not yet been clearly elucidated, is being emphasized. Many genes called sialidases are being studied and is thought that the NEU1 gene may be particularly important in the etiopathogenesis of HSP. The aim of this study is to investigate the role of the NEU1 gene in the etiopathogenesis of HSP and its relation to renal involvement. Materials and methods: Fifty patients followed in the Celal Bayar University Hafsa Sultan Hospital Pediatric Nephrology Department, with the diagnosis of HSP renal involvement were included into the study. For the control group, age and gender matched 50 cases were accepted among the outpatients admitted to Pediatric Department without any chronic diseases. NEU1 gene mutation analysis was performed in blood samples of both patient and control groups by using the Sanger DNA sequencing method. Results: NEU1 genetic mutation was not detected in any HSP patient with renal involvement and control group. Conclusion: In our study, the NEU 1 gene was not found to be associated with HSP nephritis. No changes were detected in the investigated regions of the NEU1 gene. © 2022, Pamukkale University. All rights reserved.Item Nutritional Knowledge and Ergogenic Aid Using Status of Competitive and Recreational Cyclists(Pamukkale University, 2022) Sak D.; Dayi T.; Günay E.; Öniz A.This study aimed to determine cyclists' nutritional knowledge and habits and nutritional ergogenic aid usage and shed light on the relation between cyclists' nutritional knowledge and ergogenic aid usage. It was conducted in Cyprus. There were three groups (study groups: competitive cyclists-CC, recreational cyclists-RC, and control group-CG: sedentary adults). Data were collected with a questionnaire that includes three sections (the first two sections were related to general nutritional habits, ergogenic aids, and anthropometric measurements, and the last section included The Nutrition for Sport Knowledge Questionnaire-NSKQ). All data were evaluated by Statistical Package for the Social Science-SPSS version 24.0. Totally 174 adults participated voluntarily in this study (n: 58 for each group). CC and RC skipped their main meals and smoked less than CG (p<0.05). Although most of the participants had poor sports nutrition knowledge (60.0%), CC had the highest NSKQ score, and this result showed us that CC had more information about sports nutrition than CG (p= 0.001). According to ergogenic aids usage, CC (56.9%) used more than RC (39.7%) (p= 0.000). This study showed that cyclists (CC and RC both) -who used ergogenic nutritional aids- had lower NSKQ scores (p= 0.027). In conclusion, the nutritional knowledge levels of the cyclists were found to be insufficient. It was observed that cyclists had significant differences in nutritional knowledge and low level of knowledge about ergogenic supports. Thus, continuous sports nutritional education is necessary to increase their knowledge. © 2022 The Author(s).Item The effect of salinity stress on germination parameters in Satureja thymbra L. (Lamiaceae)(Pamukkale University, 2022) Oz U.Salinity is an important problem all over the world. The destructive effect of salinity is observed from the seed germination stage. In this study, it was aimed to determine the effect of salinity on seed germination of the medically important Satureja thymbra L., whether pre-treatments are a factor in breaking the salinity stress, and to determine the level of salinity tolerance of this species. In the research, firstly, the seeds were exposed to two pre-treatments (80°C (5 minutes) + 10 ppm GA3 (24 hours), 80°C (5 minutes) + 100 ppm GA3 (24 hours)) and then 8 different NaCl concentrations (0.1 g/l, 1 g/l, 2.5 g/l, 5 g/l, 7.5 g/l,10 g/l, 15 g/l and 30 g/l) were tried. Germination seeds were counted every day and the effects of salinity on germination characteristics were investigated. The highest germination percentage (90%) was obtained at 0.1 g/l NaCl after 80°C (5 min.) + 100 ppm GA3 (24 h.) pre-treatment. The results showed that the effect of salinity was significant on germination parameters in p < 0.05. Obtained results showed that the highest NaCl concentration at which Satureja thymbra seed could germinate was 10 g/l. © 2022 Pamukkale University. All Rights Reserved.Item Frequency of rheumatic diseases in patients with familial Mediterranean fever; [Ailesel Akdeniz ateşi hastalarında romatizmal hastalıkların sıklığı](Pamukkale University, 2023) Yener G.O.; Yüksel S.; Tekin Z.E.; Türkmen H.Purpose: Mutations in the Mediterranean FeVer (MEFV) gene, which causes familial Mediterranean fever (FMF), may also cause the emergence of other specific rheumatic diseases. This study aims to determine the frequency of other rheumatologic diseases in paediatric FMF patients, evaluate whether there are clinical and genetic differences between those with and without concomitant rheumatologic diseases, and compare the data with previous studies. Materials and methods: The files of FMF patients who were followed up at the paediatric rheumatology department were reviewed retrospectively. Demographic data, MEFV mutations, treatment, disease severity scores, and concomitant rheumatic diseases were recorded from the files. Results: There were 303 FMF patients (154 female/149 male). The mean age at diagnosis was 7.04±3.9 years. The mean disease duration was 5.33±3.13 years. In the cohort, 41 FMF patients (13.5%) were diagnosed with another rheumatic disease. There were 22 cases of juvenile idiopathic arthritis (53.6%), seven cases of vasculitis (17%), six cases of periodic fever aphthous stomatitis and adenitis syndrome (14.6%), three cases of Behçet's disease (7.3%), two cases of acute rheumatic fever (4.8%), and one case of systemic lupus erythematosus (2.4%). Thirty-two of these of these 41 FMF patients (78%) had the M694V mutation (homozygous in 11, heterozygous in 21). Disease activity scores Pras and ISSF scores were higher in FMF patients with rheumatic diseases (p=0.002 and p<0.001, respectively). Conclusion: Other rheumatologic diseases should be evaluated in FMF patients. Regarding other accompanying rheumatic diseases, the M694V mutation and disease severity scores are notable factors. © 2023, Pamukkale University. All rights reserved.Item Relationship between duration of undiagnosed illness, clinical features and cognitive impairment in bipolar disorder; [Bipolar bozuklukta tanısız geçen hastalık süresinin klinik özelikler ve bilişsel bozulmayla ilişkisi](Pamukkale University, 2024) Atay E.; Aydemir Ö.Purpose: It is believed that a delay in the diagnosis of bipolar disorder may adversely affect the clinical course and outcome. This study aimed to investigate the relationship between diagnostic delay and clinical variables, as well as neurocognitive and social cognitive disorders. Materials and methods: Eighty-four patients with bipolar disorder in remission were included in the study. Participants were evaluated using a neuropsychological battery that assessed verbal memory and learning, visual memory and learning, verbal fluency, attention, processing speed, executive functions, working memory, and social cognition. Results: The duration of undiagnosed illness was longer in patients with bipolar II disorder, those without psychotic features, those with at least one suicide attempt, those whose first episode was depressive, and those currently on antidepressants. A significant positive correlation was found between the duration of undiagnosed illness and scores on the Controlled Oral Word Association Test, total number of episodes, hypomanic episodes, depressive episodes, and their respective durations. Conversely, a significant negative correlation was found between the duration of undiagnosed illness and both the number and duration of manic episodes. Conclusion: We found that a delay in diagnosis and treatment was associated with more recurrences in bipolar disorder, an increased number of depressive episodes, and at least one lifetime suicide attempt. However, the association between extended periods of untreated illness and poor clinical and functional outcomes did not align with cognitive impairment. © 2024, Pamukkale University. All rights reserved.Item Attitudes of first and sixth year medical faculty students towards sexual assault victims: a cross-sectional study; [Birinci ve altıncı sınıf tıp fakültesi öğrencilerinin cinsel saldırı mağdurlarına karşı tutumları: kesitsel bir çalışma](Pamukkale University, 2024) Turan F.; Zeybek V.Purpose: Sexual violence encompasses a range of coercive behaviors, from verbal harassment to forced penetration, and includes from social pressure and intimidation to the application of physical force. Sexual violence has numerous mental and physical health consequences on the individual. Although physicians recognize their crucial role in caring for sexual violence victims, various attitudes and behaviors hinder their fulfillment of these roles. In our study, the aim was to determine the attitudes of first and sixth year medical faculty students towards sexual assault victims and the socio-demographic factors influencing these attitudes. Materials and methods: A total of 370 voluntary first-and sixth-year students enrolled in the 2021-2022 academic year at Manisa Celal Bayar University Faculty of Medicine were administered a questionnaire comprising socio-demographic characteristics, the "Social Attitudes Scale Towards Sexual Assault Victims", and questions assessing their knowledge of forensic medicine regarding sexual assault. Results: In our study, it was found that 55.9% of the medical faculty students who participated were female, with an average age of 20.83±2.85 years. 56.5% were first-year students. 18.9% reported that they or someone close to them had been subjected to behavior considered as sexual assault. The average score of the participants in the Social Attitudes Scale towards Sexual Assault Victims was determined to be 95.29. It was statistically significant that women, those aged between 17-22, and those with a high monthly family income had higher average scores on the scale. It was found that 17.3% of the participants answered all six forensic medical questions correctly. The total scores of female students on the Social Attitudes Scale Towards Sexual Assault Victims and the scores of the behavior and thought sub-dimensions were found to be statistically significantly higher than those of male students. Conclusion: It was determined that participants generally held a positive attitude towards sexual assault victims. Factors contributing to this positive attitude included higher levels of parental education, the influence of education received during medical school and preceding periods, and social environments. © 2024, Pamukkale University. All rights reserved.Item Taxonomic characterization and secondary metabolite production of newly isolated Streptomyces sp. MC12(Pamukkale University, 2024) Oskay M.An actinobacterium newly isolated from soil during a screening study was identified as Streptomyces sp. MC12 (GenBank accession number: PP757795) based on 16S rRNA analysis. For secondary metabolite production, fermentation was carried out in ISP 2 broth at 30°C, pH 7.3, for seven days under shaking conditions at 180 rpm. As a result of fermentation studies, the antagonistic effect of the crude extract, obtained through ethyl acetate extraction, against various microorganisms was determined. The MIC values of the extract against Staphylococcus aureus and Escherichia coli were 101.3 µg/mL and 153.6 µg/mL, respectively. It was also found to exhibit strong antifungal activity against Penicillium spp. Streptomyces sp. MC12, which displays both antifungal and antibacterial properties, is considered a potential secondary metabolite producer for future studies, particularly in pharmacology and the biocontrol of fungal pathogens. © The Author(s) 2024.Item The effect of cardiometabolic control and malnutrition on the prevalence and prognosis of diabetic retinopathy in type 2 diabetes; [Tip 2 diyabette kardiyometabolik kontrol ve malnütrisyonun diyabetik retinopati prevalansı ve prognozuna etkisi](Pamukkale University, 2024) Özdemir M.; Dağ Y.; Uzunlulu M.; Oğuz A.; Ardagil A.; Acar Ö.; Selbest B.Purpose: This study aims to investigate the predictive value of cardiometabolic control, gamma glutamyl transferase (GGT) and malnutrition-related inflammation markers for predicting diabetic retinopathy (DR) prevalence and prognosis. Materials and methods: Type 2 Diabetes Mellitus patients who were consecutively admitted to Internal and Ophthalmology outpatient clinics were included in this study. Clinical, haematological and biochemical data were recorded. Cut-off values of GGT, hemoglobin, albumin, lymphocyte and platelet (HALP) score, nutritional risk index (NRI) and prognostic nutritional index (PNI) scores were determined by receiver operator characteristic curve analysis. Univariate and multivariate analyses were performed to determine the association of all variables with DR. We evaluated which of these tests were predictive and prognostic for the development of DR. Results: This study included 166 patients. Fasting blood glucose (p<0.001), creatinine (p=0.01), HbA1c (p<0.001) and microalbuminuria (p=0.01) were higher in patients with retinopathy. Mean arterial pressure (p=0.01), fasting blood glucose (p=0.03), triglyceride (p=0.008), body mass index (BMI) (p=0.02) and HbA1c (p=0.04) increased significantly as GGT level increased. Contrary to the literature, HALP, PNI and NRI scores were not associated with DR. Conclusion: Duration of diabetes, cardiometabolic control and GGT level are variables with predictive value for the prognosis of DR. No significant correlation was found between malnutrition-related inflammation markers and DR development and stage. © 2024, Pamukkale University. All rights reserved.
