Item Essential oil composition and antioxidant activities of alkanet (Alkanna tinctoria subsp. tinctoria)(2010) Ozer M.S.; Sarikurkcu C.; Tepe B.; Can S.This study is designed to examine the chemical composition and in vitro antioxidant activity of the hydrodistillated essential oil and the various extracts of alkanet (Alkanna tinctoria subsp. tinctoria). Gas chromatography (GC) and GC-mass spectrometry (GC-MS) analysis of the essential oil were resulted in the determination of 27 different compounds, representing 93.32% of the total oil. The major compounds detected in the oil, were pulegone (22.27%), 1,8-cineole (13.03%), α-terpinyl acetate (6.87%), and isophytol (6.83%), respectively. Antioxidant activities of the samples were determined by 4 different test systems namely β-carotene/linoleic acid, 1,1-diphenyl-2-picrylhydrazyl (DPPH), reducing power, and chelating effect. In all systems, essential oil showed the weakest activity profile. On the other hand, ethyl acetate and water extracts exhibited excellent antioxidant activities. As well as the antioxidant activities of the extracts, they were evaluated in terms of their total phenolic and flavonoid contents. In parallel to the experiments, ethyl acetate and water extracts were found to be rich-in these phytochemicals. © KoSFoST and Springer 2010.Item Effects of Galium aparine extract on the cell viability, cell cycle and cell death in breast cancer cell lines(Elsevier Ireland Ltd, 2016) Atmaca H.; Bozkurt E.; Cittan M.; Dilek Tepe H.Ethnopharmacological relevance Galium species have been traditionally used for its anti-cancer, antioxidant, anti-inflammatory, antimicrobial and cardioprotective effects in the folk medicine. Galium aparine (GA) is a typical climbing plant growing widespread in Anatolia. Aim of the study To investigate the potential anti-proliferative and apoptotic effect of GA methanol (MeOH) extract on MCF-7 and MDA-MB-231 human breast cancer cells and MCF-10A untransformed breast epithelial cells. Materials and methods First, the extract was characterized by both liquid chromatography/quadrupole time-of-flight mass spectrometry (LC/Q-TOF/MS) and gas chromatography-mass spectrometry (GC-MS) analyses. Then, cell viability and cell cycle distribution were investigated by XTT assay and PI staining by flow cytometry, respectively. Cell death was determined by Annexin V FITC/7-AAD staining. Results A total of 14 major phytochemicals were identified by LC/Q-TOF/MS and 34 volatile compounds were determined by GC-MS. The extract was cytotoxic in both breast cancer cell lines in a concentration and time dependent manner and showed G1 block after 72 h extract treatment. However, it was not cytotoxic to MCF-10A breast epithelial cells. Flow cytometry analyses revealed that apoptosis was induced in MDA-MB-231 cells; however, necrosis was induced in MCF-7 cells. Conclusion Our study suggests that GA MeOH extract may have potential anti-cancer effects against breast cancer cells without impairing normal breast epithelial cells. Ability to induction of non-apoptotic cell death besides apoptotic cell death by this complex plant-derived mixture may enable the killing of apoptosis resistant breast cancer cells but further studies should be conducted to investigate the bioavailability and metabolism of it in vivo. © 2016 Elsevier Ireland Ltd. All rights reserved.Item Review of the health effects of berries and their phytochemicals on the digestive and immune systems(Oxford University Press, 2018) Govers C.; Kasikci M.B.; van der Sluis A.A.; Mes J.J.Berries are generally considered beneficial to health. This health-promoting potential has mainly been ascribed to berries' phytochemical and vitamin content, and little attention has been paid to the potential benefits of berries for the digestive tract, despite this being the first point of contact. In vivo studies that described the health effects of berries on individual parts of the digestive tract (ie, the mouth, esophagus, stomach, small and large intestine, microbiome, and immune system) were reviewed. Immune effects were included because a large part of the immune system is located in the intestine. Beneficial health effects were mainly observed for whole berry extracts, not individual berry components. These effects ranged from support of the immune system and beneficial microbiota to reduction in the number and size of premalignant and malignant lesions. These results demonstrate the potency of berries and suggest berries can serve as a strong adjuvant to established treatments or therapies for a variety of gastrointestinal and immune-related illnesses. © The Author(s) 2017. All rights reserved.Item Enzyme and biological activities of the water extracts from the plants aesculus hippocastanum, olea europaea and hypericum perforatum that are used as folk remedies in Turkey(MDPI AG, 2020) Sarikurkcu C.; Locatelli M.; Tartaglia A.; Ferrone V.; Juszczak A.M.; Ozer M.S.; Tepe B.; Tomczyk M.Phenolic compounds are secondary metabolites that are found ubiquitously in plants, fruits, and vegetables. Many studies have shown that regular consumption of these compounds could have a positive effect on our health. The aim of this study was to compare the phytochemical contents of the water extracts from three different plants used as folk remedies in Turkey: Aesculus hippocastanum, Olea europaea, and Hypericum perforatum. A liquid chromatography-electrospray tandem mass spectrometry (LC-ESI-MS/MS) analysis was performed to explore the phenolic profiles. The biological activities of these extracts were also evaluated in terms of their antioxidant activities (2,2-diphenyl-1-picrylhydrazyl DPPH, 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid ABTS, Ferric Reducing Antioxidant Power Assay FRAP, cupric ion reducing antioxidant capacity CUPRAC, β-carotene, phosphomolybdenum, and metal chelating) and enzyme inhibitory properties (against acetylcholinesterase, butyrylcholinesterase, and tyrosinase). The aqueous extract of H. perforatum showed the highest levels of total phenolic, flavonoid, and saponin contents. Protocatechuic acid, vanillic acid, verbascoside, hesperidin, hyperoside, apigenin 7-hexosides, and quercetin were the most common compounds found in this species. The results confirm that A. hippocastanum, O. europaea, and H. perforatum represent a potential source of natural-derived molecules with positive properties that could be used as valid starting point for new food supplements, and drugs in the pharmaceutical, cosmetic, and food industries. © 2020 by the authors.
